The Effects of Resveratrol on Neuronal Apoptosis Induced by Hypoxia

Abstract

Objective:To explore the effects of resveratrol(RES) on neuronal apoptosis induced Hypoxia in cortical neurons.Methods: The present study established an hypoxic model in primary cultured cortical neurons of mice.In the hypoxia model,the level of ROS was determined using DCFH-DA and fluorescence microscopic imaging systems and apoptosis was analyzed by staining of propidium iodide(PI) and Hoechst 33342.Results: After pretreated with 25 μM RES for 60 min before hypoxia,the level of ROS and neuronal apoptosis were significantly lower than those in hypoxia cells.And pretreatment with 1 mM NAC(a ROS scavenger) can also reduce neuronal apoptosis percentage induced by hypoxia in neurons.Conclusion: The results indicated that RES could inhibit the hypoxia induced apoptosis in cortical neurons via reducing the level of ROS.
