Abstract
Objective To investigate effects of nuclear M-CSF on the sensitivity of anti-tumor chemotherapeutic drug in human cervical carcinoma HeLa cells. Methods Nuclear in M-CSF-transfected HeLa cells(referred as to HeLa-M cell),pCMV、cyto、myc transfected HeLa cells(referred as to HeLa-C cell) or untransfected HeLa cells(referred as to HeLa cell) were cultured in vitro.After Treating the HeLa-M cells、HeLa-C cells and HeLa cells with cisplatin(0.2 、2.0、20 μg/mL)or paclitaxel(0.2、2.0、20 μg/mL)for 48 h,the cells viability was determined using MTT assay.Western Blot was used to detect Bcl-2 expression. Results The MTT assay showed that cell viability inhibition in HeLa-M cells treated by cisplatin and paclitaxel(20 μg/mL or 2.0 μg/mL) were more than HeLa-C cell and HeLa cell(P0.01).However,at 0.2 μg/ mL there were no defference in the three kind of cells(P0.05).Nuclear M-CSF up-regulated the expression of Bcl-2 expression in HeLa cell(P0.01). Conclusion Nuclear M-CSF enhances chemotherapeutic drug resistance and apoptotic resistance of HeLa cells.
