Abstract
ObjectiveTo investigate the effect of sevoflurane on the tissue permeability of ischemia-reperfusion rat lungs.MethodsNinety-six adult male Wistar rats weighing 250~350 g were randomly divided into 4 groups:group C(n =24),which had continuous perfusion without ischemia;group IR(n =24),which had occlusion of the left pulmonary hilum for 45 min followed by reperfusion;group sev-C(n =24),which had 1MAC sevoflurane inhalation for 30 min followed by continuous perfusion;group sev-IR(n =24),which had 1MAC sevoflurane inhalation for 30min followed by ischemia-reperfusion.In each group,the indexes were measured at 45 min of ischemia and at 60 min and 120 min of reperfusion.Moreover,bronchial lavage was performed for six rats in each group at 120 min of reperfusion.MAP,SpO2,wet-to-dry weight ratio(W/D) and lung permeability index(LPI) were calculated at all time points.The changes of pulmonary morphology were observed under light and electron microscope.ResultsCompared with groups C and Sev-C,W/D and LPI increased progressively after reperfusion in rat lungs in groups IR and sev-IR(P 0.05).Moreover,compared with group IR,pre-administration of sevoflurane could inhibit the increase of W/D and LPI after reperfusion(P 0.05).In group IR,the changes of pulmonary morphology were gradually aggravated after reperfusion with disconnection of tight junctions of endothelial cells.Compared with group IR,the degree of injury in group Sev-IR was significantly ameliorated.ConclusionSevoflurane protects lungs from I/R injury by decreasing pulmonary tissue permeability.
