Abstract

Objective:To study the effects on the colony-forming capacitity of hematopoietic cells by vascular endothelial growth factor.Method:First, BALB/c bone marrow hematopoietic cells(MNCs) were prepared. Some samples were pretreated with or without hVEGF for 24 hours and then used for CFU-GM、CFU-E、CFU-S assay. Second, Others were used for CFU-GM、CFU-E assay directly while hVEGF was also added into the culture system.Result:When BALB/c bone marrow MNCs were pretreated with hVEGF for 24 hours, the numbers of CFU-GM、CFU-E、CFU-S( 33.21± 2.84、 72.00± 4.55、 12.20± 1.32 )were significantly higher than those of untreated group( 19.60± 2.10、 38.86± 2.77、 6.10± 1.52)(P 0.05). In the culture system of CFU-GM、CFU-E assays when fresh samples were used, the existence of hVEGF also promoted the number of CFU-GM、CFU-E( 73.12± 3.80 vs 51.39± 2.60, 198.98± 4.92 vs 164.40± 5.32)(P 0.05).Conclusion:Not only can hVEGF sustain but also promote the colony-forming capacity of mice bone marrow mononuclear cells.
