Abstract

Objective: To establish a stable asthma model used to screen and identify therapeutic drug for bronchial asthma.Methods: 16 SPF female BALB/c mice were randomly divided into control group and model group.The model group was sensitized by intraperitoneal injection with OVA aluminum hydroxide solution on day 0 and 14.Mice were challenged via the airways with OVA(1% in saline) for 3 days(from day 28 to 30) using ultrasonic nebulization.The control group received saline as the substitution of OVA solution.Results: Compared to the control group,the model group presented with the symptoms of acute asthma,such as scratching the head continuously,lifting the forelimbs,bowing the back,breathing deeply and fast,staying still and so on.There were more eosinophils and leukocytes in the BALF of model group.In the model mice,the inflammatory cells infiltrated around small bronchus and concomitant vessels.The smooth muscle cell proliferation,goblet cell hyperplasia and pulmonary emphysema were more serious in the model mice.Conclusion: The OVA-induced mouse model of asthma is successful.
