Callus Induction and Plant Regeneration from Seeds of Zoysia japonica

Abstract

With mature Zoysia japonica weeds explanet,studied the influence of different treatment of weeds,basic medium and the addition of growth regulator and chitosan to the induction of callus and the effect of subculturing times on the plantlet regeneration.Compared on the frequency and the forms of callus,the best method to induce callus was in N6 medium,under dark condition and with the combination of 2,4-D(4 mg/L)and chitosan(4 g/L).Under such condition,the induction frequency of callus was 100%,while the percentage of embryogenic callus was 84.4%.Regenerated plantlets with regeneration frequency 81.7% were obtained after transferring embryogenic callus to the 1/2MS medium with 2,4-D 0.1 mg/L.
