Abstract

Objective:To construct the recombinant secretory Pichia expression vector for mature peptide of human BMP-7.Methods:The cDNA encoding the mature peptide of hBMP-7was amplified by PCR and inserted into the secretory expression vector pPIC9.The recombinant was transformed into GS115after it was linearized with SalI.The transformants with phenotype His+Mut+were selected via PCR assay.Results:DNA sequencing and PCR analysis indicated that the recombinant plasmid pPIC9/BMP-7was constructed successfully and transformants were screened out for expression.Conclusion:The constrnction recombinant plasmid pPIC9/BMP-7provided a basis for expression of mature peptide of hBMP-7by geˉnetic engineering.
