Abstract
Objective To explore the effects of fluoride and aluminum on mRNA expression of OPG and RANKL in MC3T3-E1,and mouse osteoblastic cell line.Methods Cells were cultured in medium contained 50 μmol/L NaF or/and 5 μmol/L AlCl3.Expressions of OPG mRNA and RANKL mRNA were determined and carried out semiquantitative analysis by RT-PCR 72 h later.Results Compared with that of control group,expressions of OPG mRNA in MC3T3-E1 treated with fluoride and aluminum were increased significantly(P0.05),fluoride and aluminum had synergistic effect on expressions of OPG mRNA in MC3T3-E1 cells(P0.05).However,the expression of RANKL mRNA showed no obvious change among different groups.Therefore,the ratio of RANKL/OPG was significantly decreased(P0.05).Conclusion The results suggested that the combined administration of fluoride and aluminum might inhibit the differentiation and maturation of osteoblast by stimulating expression of OPG mRNA;the decreased ratio of RANKL/OPG indicated that the process of bone formation would be greater than that of bone absorption.
