Abstract
Objective:To investigate the function of porcine islet cells ameliorating the diabetic mice hyperglycemia.Methods:The diabetic mice were made by injecting STZ into the abdominal cavity. The porcine islets were seperated by collagenase and purified by density gradient centrifugation with Ficoll liquid and transplanted into the diabetic mice abdominal cavity. The blood glucose was measared after transplantation to observe the effects. Results: On the 3rd day after transplantation the blood glucose began decrearing obviously. The effective existing time of porcine istet cells was 5~12 days,and the average surviving time of graft was 7.1±1.87 days.The blood glucose in transplanting group decreased to 5.77±1.38 mmol/L,while in control gronp was 26.19±3.05mmol/L.Conclusions: The porcine islet cells can ameliorate the hyperglycemia of diabetic mice effectively,and transplantation of xenograft has curing effects to diabetic mice.
