EFFECTS OF HO-1 ON DOPAMINERGIC NEURONS IN MODEL MOUSE OF PARKINSON DISEASE

Abstract

Objective To investigate the effect of inhibition of heme oxgenase-1(HO-1) on dopaminergic neurons in the substantia nigra(SN) in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-induced mouse model of Parkionson disease.Methods C57BL / 6mice were divided into three groups(control group,MPTP group and ZnPPIX+MPTP group) in random,with six in each group.A mouse model of Parkinson ’ s disease was created by i.p.injection of MPTP(30mg / kg.),once daily for five days.The mice in the MPTP group were given normal saline(2 μ L,i.c.v.) 4hbefore MPTP injection-for eight days;those in ZnPPIX+MPTP group,microinjection of ZnPPIX(50 μ g / kg) was given,4hbefore the injection of MPTP,for eight days;for those in the control,MPTP and ZnPPIX were replaced by normal saline.The expression of HO-1and TH in the SN was assessed by western blots.Results The expression of HO-1in mouse SN of ZnPPIX+MPTP was lower than that of the control group and MPTP group(F=6.844;q=5.943,4.444;P0.05).In MPTP group and ZnPPIX+MPTP group,the TH expression in SN area was lower than that in the control(F=5.095;q=4.303,4.030;P0.05),the difference of TH expression between ZnPPIX +MPTP and MPTP group was not significant(q=0.234,P0.05).Conclusion Inhibiting the expression of HO-1does not play a significant role in deteriorating the degeneration of dopaminergic neurons of Parkinson ’ s mouse induced by MPTP.
