Separation and purification of total alkaloids from Zanthoxylum nitidum(Roxb.)DC.with D-101 macroporous resin

Abstract

Objective To explore a process to separate and purify total alkaloids from Zanthoxylum nitidum(Roxb.)DC.with macroporous resin.Methods UV was used to determine the total alkaloids while TLC was used to analyze the separation efficacy.Results Extract of volume fraction 60% alcohol-5 g·L-1 hydrochloric acid was absorbed and separated with D-101 macroporous resin.Then the alcohol in the effluent was recycled and reduced to 45%,and the concentrated solution was absorbed and separated with the resin.Finally,the alcohol in the effluent was recycled and reduced to no alcohol flavor,and the concentrated solution was absorbed and separated with the resin.Total alkaloids were eluted with 95%,80% and 50% alcohol,respectively.Mass fraction of total alkaloids in products was 18.07%-27.67%.Conclusion The process not only enriches total alkaloids,but also separates simply ingredients with different polarity.
