Effects of Mosapride on Gastric Emptying and Plasma Motilin in Oral Rehydration of Rats with 40% Blood Volume Loss

Abstract

Objective To investigate the effects of mosapride on gastric emptying and plasma motilin in oral medication of glucose electrolyte solution(GES) in hemorrhagic rats.Methods SD rats were randomly divided into GES group(oral GES after sham operations),H+GES(oral GES after hemorrhage),and GES/MOS(oral GES and mosapride).About 40% of total blood volume was let from rat carotid arteries to establish models of hemorrhagic shock in all groups.GES were given GES lavage by doubling the blood loss at min 30,60,respectively.Mosapride(0.5 mg·kg-1·w·-1)was dissolved and given in the first intragastric administration.The gastric emptying rate was determined by phenol red method and plasma motilin by radioimmunity at hour 4 after hemorrhage.Results The gastric emptying rates of groups GES,H+GES and H+GES/MOS were(92.4.3±12.5)%,(62.4±11.4)% and(79.6±10.7)%,respectively,at hour 4 after hemorrhage.There was significant difference between 3 groups(P0.05).The plasma motilin of H+GES/MOS group was dramatically higher than that of H+GES(103.8±11.4 vs.72.5±14.3,P0.05),and lower than that of GES(132.3.8±24.1,P0.01)at hour 4 after hemorrhage.The plasma NO content was significantly lower in H+GES/MOS group than in H+ GES(83.2±12.6 vs.105.7±13.2,P0.01),but still higher than in GES(53.6±6.9,P0.05).Conclusion Mosapride can increase significantly plasma motilin levels,reduce NO,promote the stomach to evacuate oral solution and enhance the effects of oral rehydration in rats with hemorrhagic shock.
