Abstract
Objective To investigate the sensitivity changes of hepatoma cell to ciglitazone after the activity of NF-κB was inhibited with NF-κB decoy oligodeoxynucleotides(ODNs).Methods NF-κB decoy ODNs were transfected into HepG2 cells with lipofectamine,the activity of NF-κB was determined before and after transfection,respectively.The transfected HepG2 cell was treated with ciglitazone and then the growth was recorded with growth curve,the cell cycle was detected with by flow cytometry,the level change of Cyclin D1 was determined with Western blot.Results The activity of NF-κB in transfected HepG2 cells was significantly decreased;the inhibitory effect of ciglitazone on the proliferation of HepG2 cell was enhanced,70.65% cells was arrested in G1/G0 phase,the expression of Cyclin D1 was obviously decreased.Conclusion NF-κB decoy ODNs could improve the sensitivity of HepG2 cells to ciglitazone,and the effect may relate with the downregulation of NF-κB activity,enhancing the inhibitory effect on the proliferation of hepatoma cells and arresting the cell cycle.
