Abstract
Objective To explore the proper cell seeding density and co-culture time of the bone marrow stromal cells (BMSCs)-coral construct. Methods BMSCs were isolated and cultured in osteogenic-induced and non-induced medium. The induced cells were complex with coral to co-culture, non-induced BMSCs-coral construct was served as control. The constructs were examined from adherent rate, growth curve, Scan electron microscopy (SEM) and quantitative histochemistry. Results Quantitative histochemistry demonstrated the higher alkaline phosphatase (AKP) and osteocalcin expression of induced BMSCs-scaffold than those of non-induced BMSCs-scaffold. The adherent rate significantly decreased when the cell seeding density exceeded 1.5×107/cm3. The growth curve demonstrated the proliferation of BMSCs on coral reached plateau after 7 days co-culture. Also SEM and quantitative histochemistry showed BMSCs grew well and began to express osteocalcin on the coral after 7 days. Conclusions The osteogenic ability was higher of induced BMSCs-scaffold than non-induced BMSCs-scaffold. The proper seeding density and co-culture time of the construct were 1.5×107/cm3 and 7 days respectively.
