Abstract

Objective To explore the influence of IL-4 on the expression of TNF- α,IL- 1β,IL- 6 and MMP9 in LPS- activated murine microglia cells. Methods LPS- activated microglia cells were treated in vitro for 24 h in the presence of IL- 4. The levels of TNF- α, IL- 1β,IL- 6 and MMP9 in the supernatant of culture were measured by enzyme- linked immunosorbent assay( ELISA),and the expression on mRNA level of TNF- α,IL- 1β,IL- 6 and MMP9 had been detected by RT- PCR. Results The concentrations of TNF- α,IL- 1β,IL- 6 and MMP9 in the supernatant of culture and mRNA levels of TNF- α,IL- 1β,IL- 6 and MMP9 were evidently increased in LPS- activated microglia cells and they were prominently decreased by co- incubation with IL- 4. Conclusion According to this study in mice,IL- 4 shows neuroprotective function by inhibiting the expression of inflammatory related factors in activated microglia cells.
