Effect of atorvastatin on cardiac inflammation in rat with type 2 diabetic

Abstract

Objective:To study the effect of atorvastatin on cardiac expression of NF-κB,TNF-α,MMP-9 in rats with type 2 diabetic mellitus(DM).Method:Sixty SD rats were randomly divided into 3 groups: control group(N group),DM group(D group) and atorvastatin group(A group).Plasma and cardiac tissue TNF-α were assessed by RIA.RT-PCR was used to study the gene expression of NF-κB,TNF-α and MMP-9.The proteins of NF-κB,TNF-α,MMP-9 were evaluated by immunohistochemical method.Result:Compared with the N group,plasma and cardiac tissue TNF-α in D group increased significantly(P0.01),while gene and protein expression of NF-κB,TNF-α,MMP-9 in myocardium of rat with DM increased significantly(P0.01).But atorvastatin can reduce the level of TNF-α,NF-κB,MMP-9.Conclusion:Expression of NF-κB,TNF-α,MMP-9 were decrease in serum or myocardium after treatment of atorvastatin in rats with type 2 DM,suggesting that atorvastatin could inhibit the inflammation of myocardium with DM.
