Abstract
Objective Survivin short hairpin RNA(shRNA) recombinant plasmid vectors were transfected to B cell lymphoma Raji cells.Influence of survivin gene expression and growth inhibition of Raji cells induced by survivin expression vectors was observed in vitro and in vivo.Methods The experiments were divided into four groups of blank control(D),A,B and C.The stable Raji cells transfected with pGenesil-1-survivin-A(group A),pGenesil-1-survivin-B(group B) and pGenesil-1-NC(group C,negative control) were obtained by G418 selection.The expressions of survivin mRNA and protein were estimated by RT-PCR and FCM.Cell proliferation ability was evaluated by cell count.Results Stable Raji cells transfected with various recombinant plasmids were successfully screened by G418.The inhibition rates in groups of A and B were 72.71% and 45.50% for survivin mRNA and 64.15% and 41.16% for survivin protein,respectively,(P0.05).The growth abilities of those two stable transfection Raji cells were significantly decreased.The interference ability was stronger in group A than that in group B.So did the inhibitory effect of tumor cell proliferation.Conclusion Two recombinant plasmids of pGenesil-1-survivin-A and pGenesil-1-survivin-B can interfere the survivin gene of Raji cells and inhibit the proliferation ability of Raji cells.
