Abstract
Objective:To detect the reasons for HbsAg false negative in one-step ELISA assay.Methods:Results of serum samples with HBsAg(-)、HBsAb(-)、HBeAg(+)、HBeAb(-)、HBcAb(+) in one-step ELISA assay were reexamined.HbsAg of these samples were both detected in an ELISA two-step sandwich assay and multiproportion-dilution one-step ELISA assay.Results:50 out of 15 809 samples were found with HBsAg(-)、HBsAb(-)、HBeAg(+)、HBeAb(-)、HBcAb(+) in one-step ELISA assay,account a 0.32% proportion.5 samples of all these turned out to be positive in both ELISA two-step sandwich assay and the multiproportion-dilution one-step ELISA assay.Conclusion:False negative does exist in a high-dose HBsAg sample due to the hook effect.Clinicals can choose either ELISA two-step sandwich assay or a multiproportion-dilution one-step ELISA assay to avoid this problem.For the low-dose ones,a higher sensitive method should be developed.
