Abstract
Objective To study the biological behavior of CD147 in prostate cancer cellline PC-3,and establish the lower expression of CD147 stable prostate cancer line PC-3 by RNA interferenc.Methods Targeting CD147 interference plasmid was transfected into prostate cancer PC-3 cell by the method of lipofectamine 2000.The single cell clones from limiting dilution were screened by G418 and the mRNA and protein expression were examined by RT-PCR and Western blot.Results Compared with cells transfected pSilencer-Scramble,cells transfected pSilencer-CD147 expression vector were inhibited both mRNA and protein expression of CD147(P0.05).Conclusion Sequence-specific RNAi vector targeting CD147 is capable of suppression CD147 gene and protein expression in prostate cancer PC3 cell,which is the foundation for researching CD147 function.
