Abstract

Objective:To evaluate the function of ursolic acid on the proliferation of human Hepatoma carcinoma cells SMMC-7721.And to study the effect of ursolic acid on the apoptosis of SMMC-7721 by flow cytometry.Methods:Human hepatoma SMMC-7721 cells were co-cultured with different doses of ursolic acid for 24 h,72 h,respectively.The absorbance value was evaluated by neutral red staining method.The influence of ursolic acid on the proliferation of hepatoma cells was evaluated;and the apoptosis was detected by using flow cytometry.Results:Ursolic acid on the proliferation of SMMC-7721 cells had obvious inhibition effect(P 0.05 or P 0.01) and presented time-dose related manner.The half inhibitory concentration(IC50) of ursolic acid on SMMC-7721 cells strain was 12.59 μg·mL-1and 8.32 μg·mL-1,respectively.The result indicated that the apoptosis rate was up to 14.07% and there was a positive correlation between the drug concentration and the apoptosis rate.Conclusion:Ursolic acid on the proliferation of hepatoma SMMC-7721 cell line has the obvious inhibitory action.The inhibition may be related to the induction of apoptosis of hepatoma cells.
