Abstract

ObjectiveTo investigate expression of transforming growth factor-β_1(TGF-β_1),Smad4 and Smad7 in kidney tissiue of diabetic rats. MethodsHealthy male Wistar rats were divided into two groups randomly,normal control group and strepotozocin induced diabetic rats group(12 rats in each group).The rats were sacrificed after drug administration for eight weeks.Mean arterial pressure,plasma glucose,kidney function,24 h urinary protein were observed.The level of TGF-β_1,Smad4 and Smad7mRNA were examined by reverse transcription-polymerase chain reaction(RT-PCR). The localization TGF-β_1, Smad4 and Smad7 protein were examined by immunohistochemistry staining. ResultsCompared to normal control group, mean arterial pressure,plasma glucose,kidney function,24 h urinary protein were significantly increased in diabetic rats group. The levels of TGF-β_1 and Smad4,Smad7mRNA were up-regulated as well as the expression of TGF-β_1 and Smad4,Smad7 protein. ConclusionTGF-β/Smad signaling is activated in diabetic nephrology.
