Inhibitory effect of all-trans retinoic acid enhances on proliferation and induction of apoptosis of lung cancer cell line A549 cells in vitro

Abstract

OBJECTIVE:To investigate the inhibition effects of all-trans retinoic acid (ATRA) with cisplatin (DDP) on proliferation and apoptosis of pulmonary carcinoma cell line A549 in vitro.METHODS:ATRA and DDP were added into cultured pulmonary carcinoma cell line A549 in different ways.After 24 hours,MTT was used to determine the combinative effect and fluorogenic quantitative RT-PCR were used to examine the expression levels of RARβ mRNA and VEGF mRNA.RESULTS:All of A549 cells treated respectively with 5,10 and 50 mg/L of cisplatin showed the apoptosis in a concentration-dependent pattern (P0.05); compared to individual drug of DDP,DDP combined with ATRA could more obviously inhibite the proliferation of A549 cells,promoted the expression of RARβ mRNA while inhibited the expression of VEGF mRNA that increased the apotosis rate of the cells(P0.05).CONCLUSION:DDP combined with ATRA treatments in chemotherapy can synergistically inhibit the proliferation of A549 cells,and significantly increase the chemosensitivity on human non-small cell lung cancer.
