Abstract
Objective:To determine the potential role of apoptosis and Fas/Fas ligand system expression in the pathogenesis of acute lung injury (ALI).Methods:Using TUNEL,in situ hybridization,reversetranscription polymerase chain reaction (RTPCR),and immunocytochemistry techniques,pulmonary apoptosis, Fas/FasL expressions were detected in rats during early phase of ALI.Results:Endotoxin challenge resulted in the rapid appearance of apoptosis in alveolar epithelial cells and pulmonary vascular endothelial cells at early stage of ALI.The expression of Fas/FasL mRNA and protein was upregulated in lung tissues of rats with ALI ( P 0 01 ),which was related to the elevation of pulmonary apoptosis.Conclusions:These results suggest that the excessive apoptosis and Fas/FasL system expression may play a role in the pathogenesis of endotoxininduced ALI in rats.
