Acquiring and Analysis of 149 ESTs of Schistosoma Japonicum

Abstract

Objective To acquire and analyze adult stage Schistosoma japonicum (Chinese strain) expressed sequence tags (EST) from an adult Schistosoma japonicum cDNA library and to search new vaccine candidates and drug targets for preventing, controlling and treating schistosomiasis. Methods A cDNA library was constructed from Shistosoma japonicum (Chinese strain) adult stage. Positive clones were selected randomly from the cDNA library and were sequenced. After analyzing in GenBank databases by BLAST programs and other programs ESTs were acquired and submitted to GenBank. Then they were given accession numbers. Results 149 ESTs were acquired from a total 382 clones that were randomly selected from the adult Schistosoma japonicum cDNA library. All ESTs were successfully submitted to the dbEST at Genbank. ESTs accession numbers were CA747382~CA747391, BU917055~BU917058, CA305307~CA305309, BU581980, BU582400~BU582403, BU666906 and CA946548~ CA946666. Some of ESTs were homologous with the sequences of male, female, egg, cercariamiracidia, and schistosomula of Schistosoma japonicum. Conclusions EST strategy is a rapid, efficient and economical method to acquire the ESTs of the adult stage Schistosoma japonicum (Chinese strain) from cDNA libraries.
