Abstract

Objective To observe the expression of mGluR5 mRNA and the dynamic regulation in the cortex of rats in focal cerebral ischemia in different phases,and to investigate the significance of it in acute ischemia.Methods Fifty-five male Wistar rats were randomly separated into 11 groups:normal,sham and ischemic for 1,3,6,12 and 24 hour respectively by middle cerebral artery occlusion model.Brain The changes in mGluR5 mRNA expression and pathology of cortex were observed under light microscope with digoxin labeled oligonucleotide probe in situ hybridization method and computer aided image analysis.Results Compared with the normal and each sham control group,the expression of mGluR5 mRNA was elevated from 1h and peaked within 6h after ischemia.However,it was no significance between normal and sham control groups.Conclusions It was suggested that up-regulated of mGluR5 might be related with focal cerebral ischemia and it promoted the development of cerebrovascular disease.
