Effect of survivin gene to the expression of heme oxygenase-1 and cell apoptosis in severe acute pancreatitis associated lung injury in Wistar rats

Abstract

Objective To investigate the effects of survivin gene to the expression of heme oxygenase-1and cell apoptosis in severe acute pancreatitis(SAP)associated lung injury in Wistar rats.Methods The study enrolled 30 Wistar rats,after being randomized into groups,SAP group was established with 24 rats into four groups of 6h,12 h,24h,48h(6rats a group),and 6rats in control group.The expression of survivin gene was detected by RT-PCR,the expression of heme oxygenase-1 was tested with immunohistochemistry,TUNEL method was used to detect the apoptosis in lung tissue.Results Survivin gene and heme oxygenase-1expression in SAP group at different time were significantly higher than those of control group(P0.05).Cell apoptosis:the control group with normal structure of alveolar cells only a few part-time cells and epithelial cells apoptosis,apoptotic cells in SAP group were obvious,and increased with the time.Conclusion Survivin gene in SAP associated lung injury increased significantly,while the heme oxygenase-1expression increased,at the same time as the survivin gene expression level rises,the apoptotic cells increase.The data suggest that the increase of active survivin gene in SAP associated lung injury can cause lung injury.
