Abstract
Objective:To compare short-term response and toxicity with FOLFOX4 regimen versus leucovorin,fluorouracil and oxaliplatin regimen for treatment of advanced colorectal cancer.Methods:Total of 66 patients proved pathologically were divided into 2 groups according to the admission date,35 cases received the FOLFOX4 regimen [oxaliolatin 85 mg/m2as a 2-hour infusion on day 1 and a 2-hour infusion of LV(200 mg·m-2·d-1) followed by a 5-Fu bolus(400 mg·m-2·d-1) and 22-hour infusion(600 mg·m-2·d-1) for 2 consecutive days every 2 weeks].Leucovorin,fluorouracil and oxaliplatin regimen[oxaliolatin 130mg/m2as a 2-hour infusion on day 1,a 2-hour infusion of LV(200 mg·m-2·d-1) followed by 2~6 hours infusion(600 mg·m-2·d-1) on day 1~5 days every 3 weeks] was applied in the other group including 31 cases.Efficacy was evaluated at 2 cycles(8weeks).Results:In 35 cases with FOLFOX4 regimen,overall response rate was 51.43%(3 complete responses and 15 partial responses).Meanwhile in the other group,1 complete responses and 12 partial responses,overall response rate was 41.94%.No significant differences were found between two groups in overall response rate.The major toxicity was reversible peripheral sensory neuropathy,bone marrow suppression and nausea vomiting,no death correlated chemotherapy.Conclusion:FOLFOX4 regimen and LFP regimen are effec tive regimens.Nevertheless,overall response rate of FOLFOX4 is higher than LFP.Toxicity is well-tolerated.So,FOLFOX4 seemed beneficial as first-line therapy in advanced colorectal cancer.The aged cases may choose LFP regimen.
