Comparative pharmacokinetics of huperzine A sustained-release patches and tablets in normal volunteers

Abstract

AIM To compare the pharmacokinetics of huperzine A sustained-release patches and tablets in healthy normal volunteers.METHODS Twenty healthy volunteers were divided into two groups(n = 10 for each group),and respectively received a single dose of sustained-release patches 10 mg and tablets of huperzine A 0.2 mg.The concentrations of huperzine A were determined by LC-MS/MS method and the pharmacokinetic parameters of huperzine A were calculated by DAS software.RESULTS The pharmacokinetic parameters of the sustained-release patches and the tablets of huperzine A were as follow: tmax(40.2 ± 13.2) and(0.6 ± 0.3) h,t1/2(28.4 ± 7.4) and(13.6 ± 1.3) h,ρmax(0.7 ± 0.4) and(1.9 ± 0.5) μg·L-1,MRT0-t(74.1 ± 8.5) and(18.7 ± 1.6) h,AUC0-t(70.4 ± 53.1)(0-168 h) and(19.9 ± 2.7)(0-48 h) μg·h·L-1.CONCLUSION The method is accuracy,sensitivity,and reliability for the determination of huperzine A in plasma.Compared with the reference preparation,sustained-release patches of huperzine A has the sustained-release characteristics.
