Abstract

To investigate the effects of astrocytes and the formation of tight junctions of endothelial cells in blood-brain barrier. Immortalized endothelial cell line ECV304 was cocultured with astrocytes. The presence of tight junctions between endothelial cells was investigated by electrical microscope following silver staining. Transendothelial electrical resistance (TER) of monolayer of ECV304 was measured by Millipore-ERS system to determine the function of tight junctions. Astrocytes could induce tight junctions between endothelial cells. The TER value was increased to 321. Sficm2 after being cocultured with astrocytes. Astrocytes can induce the formation of tight junctions between endothelial cells. At the same time, this coculture method of ECV304 and astrocytes can be a useful model to investigate the molecular mechanism of the formation of tight junctions in blood-brain barrier.
