Role of TRPV1on the rats with visceral hypersensitivity induced by transient intestinal infection

Abstract

[Objective]To investigate the role of TRPV1 on the formation of visceral hypersensitivity on rats induced by transient intestinal infection.[Methods]Thirty male Sprague-Dawley rats were randomly divided into normal control,sham operation and visceral hypersensitivity.The rats in visceral hypersensitivity were infected by administering 1.0ml of PBS containing 8 000 T.spiralis larvae by gavage.Those rats in sham operation were only administered 1.0ml of PBS.Electromyography(EMG)was recorded at8 th week after adaptive feeding for evaluating visceral sensitivity.The areas under curve(AUC)of EMG in 20 swere calculated to evaluate the visceral sensitivity.Western-blot was respectively employed to detect protein TRPV1 expression.[Results]1 Under different grades levels of CRD pressure,AUCs showed significant correlation in the three groups respectively(P0.05).The coefficient of correlation and P value of the three groups were 0.825,P=0.000;0.831,0.000;0.625,0.006 respectively.Under 20 mmHg,40mmHg and 60 mmHg,the difference of the AUC among the three groups was significant(P=0.034,0.043,0.048respectively).Under 80 mmHg,no significant difference was found in three groups(P0.05).2In normal control,sham operation and visceral hypersensitivity group,the IODs of TRPV1 protein were 0.95±0.17,0.937±0.24,1.57±0.31 respectively.The expression of TRPV1 of visceral hypersensitivity was significantly increased than sham operation and normal control(P0.05).[Conclusion]The increased expression of TRPV1 plays an important role in the formation of visceral hypersensitivity.
