Mixture of low concentration dimthyl sulfoxide and second-messenger effector as cryopreserving solution in protection of platelets activity

Abstract

Objective:To study the in vitro activity of platelets following cryopreservation with mixed solution of low concentration DMSO(2%) and second-messenger effectors. Methods: Fresh platelet concentrate was cryopreserved with 6% DMSO(control group) and mixed solution of 2% DMSO and second-messenger effector(treated group) for one week. The platelet count, adrenaline and thrombin induced aggregation, and expression level of CD41,CD42b ,CD62p and CD35 were measured in the thawed platelets.Results: Compared with the control group, treated platelets had higher platelet count (P 0.05) and thrombin induced aggregation (P0.01), though still lower than fresh platelets (P0.01). Compared with fresh platelets, cryopreserved platelets showed a significant increase in CD62p expression (P0.05,P0.01), but the increase of treated platelets were significantly lower than that of control (P0.05).Adrenaline induced aggregation and expression level of CD42b of cryopreserved platelets were lower than those of fresh platelets (P0.05,P0.01),and there were no significant difference between the control and treated platelets.Conclusion: The in vitro activity of platelets cryopreserved with mixed solution 2% DMSO and second-messenger effector is greatly improved compared with platelets cryopreserved with 6% DMSO.
