Inhibitory effect of Ardipusillide on human cervical carcinoma cells

Abstract

Objective To observe the inhibitory effect of Ardipusillide on human cervical carcinoma cells (Hela cells). Methods Ardipusillide at 0.25 mg/L was added to the culture medium of Hela cells and cocultured for 0-72 h. The cells were observed at different time points. Apoptosis was detected with light microscope and DNA electrophoresis, while [Ca 2+ ]i was determined with confocal laser scanning microscopy. Results Changes of early apoptosis were found in Hela cells after treated with Ardipusillide for 24 h, and DNA ladder was found in 60 and 72 h samples [Ca 2+ ]i increased and sustained at arather high level after induced with Ardipusilliae for 60 and 72 h. Conclusion Ardipusillide has an inhibitory effect on Hela cells in vitro and could induce apoptosis. At different time points, intracellular free Ca2+ concentrations are maintained at a sustained increase.
