Abstract

【Objective】 To explore the role and mechanism of tetrahydroxystilbene-2-O-β-D-glucoside(TSG) on the apoptosis of HUVECs induced by H2O2.【Methods】 The models of apoptosis induced by H2O2 in HUVECs and the concentrations of TSG on apoptosis model were established via MTT,Hoechst33258 dyeing,and Flow Cytometry.The expression of Peroxynitrite-Poly(ADP-Ribose) Polymerase(PARP) was detected by RT-PCR and Western blotting respectively.【Results】 All of the different concentrations of H2O2 can decrease the rate of cell proliferation and increase the rate of apoptosis.Compared with H2O2(300μmol/L) group,the cell proliferation rate was no significantly difference after pretreated TSG(0.1,1μmol/L)(P 0.05) and the cell proliferation rate was increased after pretreated TSG(10,100μmol/L)(P 0.01) respectively.TSG can decrease the rate of apoptosis which induced by 300μmol/L H2O2,and the difference was significantly(P 0.01) except for 1μmol/LTSG.The expressions of PARP mRNA and protein were lower in TSG(10μmol/L) group than in H2O2(300μmol/L) group(P 0.01).【Conclusion】 TSG can inhibit the apoptosis of HUVECs induced by H2O2,and its mechanism may relate to down-regulation the expression of PARP.
