Abstract

Objective To explore a method of inducing U937 cell line to express CD14 by VitD3 and the response of U937 cells to LPS . Methods The U937 cells was cocultured with 0.1μMol VitD3 for 24h to produce CD14,which and its mRNA was observed by immunohistochemical method and in situ hybridization.The TNF-α mRNA level was detected by in situ hybridization too. The response of U937 cells to different concentration of LPS after different stimulating time was observed. Results The U937 cells could stably express CD14 and were much more sensitive to the stimulation of LPS after being induced by VitD3. Conclusions U937 cell line were ideal cell model for research on the activation of macrophages mediated by LPS because of its ability of expression of CD14 induced by VitD3.
