Abstract
Objective To set up the simultaneous genotyping of human ABO antigens by PCR-SSP assay,and to study on ABO blood group.Methods 13 pairs of sequence-specific primers were designed and synthesized.The annealing temperature for all the primer pairs,the concentrations of each primer pair and Mg~(2+) were adjusted to the optimum so that ABO alleles could be amplified simultaneously under the same PCR cycling parameters.The electrophoresis products were conducted simultaneously on the same agarose gel.34 samples of predetermined ABO phenotypes and 20 samples of A2 subgroups previously sequenced were genotyped by the method.Results A concordance rate of 100 percent was observed between the results of 54 samples by PCR-SSP and that of known common ABO phenotypes and rare A2 alleles.Conclusion ABO genotyping is appropriate for the routine ABO genotyping and research for Chinese population.
