Abstract
Objective:To investigate the influence of IL-1β,IL-6 and IL-23 on the Th17 cells formation in primary biliary cirrhosis(PBC) patients,and to compare Th17 differentiation in normal control and PBC patients.Methods:The nave T cells from the peripheral blood of normol control and PBC patients were isolated.The cells were cultured with anti-CD3,anti-CD28,anti-IL-4,and anti-IFN-γ as the basal conditions,and grouped by culturing with different cytokines to induce the differentiation of Th17 cells.The groups were ①without additional cytokines as the control group;②IL-1β group;③IL-1β+IL-6 group;and ④IL-1β+IL-6+IL-23 group.The efficiency for differentiation of Th17 cells was assessed by flow cytometry(FCM) after cultured.Results:When compared with that of the normal control,the frequency of differentiation into Th17 cells of PBC patients was the highest in the IL-1β+IL-6+IL-23 group,and the statistically difference was significant(P0.01).To the PBC patients,IL-1β group;IL-1β+IL-6 group;IL-1β+IL-6+IL-23 group could induce stronger Th17 cell differentiation significantly(P0.01);and the frequency of Th17 cell differentiation was the highest in the IL-1β+IL-6+IL-23 group.The statistical difference was significant when compared with those of the other groups(P0.01).Conclusion:IL-1β alone and combinating with IL-6 and IL-23 could induce Th17 cell differentiation in PBC patients.The inducing efficiency is significantly higher than in normal control.Combination of IL-1β,IL-6 and IL-23 in PBC patients could induce efficient Th17 cell differentiation in vitro to achieve the best.
