Abstract
Objective: To establish rapidly recurring hippocampal kindlingmodels in rats. To investigate the mechanism of the models primarily. Methods:Rapidly recurring hippocampal kindling models were established in differentparameter. The effect of general anti-kindled drugs such as diazepam andphenobarbital were observed on rapidly recurring hippocampal kindling models inrats. The number of c-fos immune-positive cells in CA3 of hippocampus andspecific cerebral cortex immunohistochemistrily in rapidly recurring hippocampalkindling and amygdala kindling models were calculated. Applying CHX 2h before thekindling stimulus, calculate the number of c-fos immune-positive cells in the samezone. Results: Rapidly recurring hippocampal kindling models were establishedsuccessfully. We found that the kindled ratio was decided by stimulus parameters. Phenobarbital at doses of 20, 50mg·kg~(-1) ip and diazepam at doses of 2, 5mg·kg~(-1) ip, can decrease the Racine's stages in rapidly recurring hippocampal kindled rats (P0.05). The number of c-fos immune-positive cells in CA_3 of hippocampus and specific cerebral cortex increased in rapidly recurring hippocampal kindling and amygdala kindling rats (P0.05). The number of c-fos immune-positive cells in the same zone in rapidly recurring hippocampal kindled rats applied CHX decreased (P0.05). Conclusion: Rapidly recurring hippocampal kindling models were established successfully. We found that the kindled ratio was decided by stimulus parameters. The number of c-fos immune-positive cells in different zone of rapidly recurring hippocampal kindling model increased.
