Comparing the application of two diluents in seminal routine examinations

Abstract

Objective:To explore diluents of sperm,with which to dilute high density sperm,the status of the sperm qualities could be reflected more literally when assayed through computer-aided sperm analysis(CASA).Methods:Total 64 shares sperm with high density(34 shares sperm were not melicera and 30 shares sperm were melicera)were all diluted at same ratio with normal saline and autologous seminal plasma respectively.Then,every patient′s original sperm and the two dilutions were all analyzed through CASA,and the dilution of normal sanline were counted the density and the motility by artificial method with Makler counting chamber.Results:In not melicera samples:by CASA,the density of original sperm was significantly higher than that of the dilution of autologous seminal plasma(P=0.003) or nomal saline(P=0.001),densities of the two dilutions were similar(P=0.076);the motility of original sperm was significantly lower than that of the dilution of autologous seminal plasma(P=0.001) or normal saline(P0.001),the motility of normal saline-diluted sperm was significantly higher than that of autologous seminal plasma(P0.001).the density(P=0.372)and the motility(P=0.060) of normal saline-diluted sample by CASA were both similar to those of the dilution by the artificial method.In melicera samples: by CASA,the density of original sperm and that of the dilution of autologous seminal plasma were similar(P=0.054),densities of them were both significantly higher than that of normal saline-diluted sperm(P0.001);original sperm and autologous seminal plasma-diluted sperm had similar motility(P=0.288),the motilities of them were both significantly lower than that of normal saline-diluted sperm(P0.001);the density(P=0.073) and the motility(P=0.161) of normal saline-diluted sperm by CASA were both similar to those of the dilution by the artificial method.Conclusions:During seminal routine examinations by CASA,the parameters of sperm diluted by autologous seminal plasma reflect the qualities of patients′ sperm more literally in the case of high density and not melicera samples;for high density and melicera sperm,normal saline is a relatively better diluent.
