Abstract
Objective To study the effect of sodium arsenite(NaAsO2) on the phosphorylated protein kinase B(p-Akt) and its downstream signaling factor I-Kappa B(IκB) and nuclear transcriptional factor Kappa B(NF-κB) in human keratinocyte(HaCat).Methods HaCat cells were exposed to sodium arsenite at the doses of 0(control group)-100 μmol/L for six hours.Then cell viability were detected by Alamar Blue assay,while the protein expression levels of p-Akt,phosphorylated IκB(p-IκB) and NF-κB were detected by Western Blot.Results The protein expression levels of p-Akt,p-IκB and NF-κB in nuclear in exposed groups were significantly higher than those in control;The cell viability and the protein expression of NF-κB in cytoplasm in exposed groups were markedly lower than those in control(P0.05).The protein expression levels of p-Akt,p-IκB and NF-κB in the nuclear of HaCat cells were gradually increased with the increasing of exposure doses,while the cell viability and the protein expression levels of NF-κB in the cytoplasm showed a decreasing trend.Conclusion NaAsO2 exposure may decrease HaCat cell viability and induce the expression of p-Akt protein up-regulation in HaCat cells,which results in phosphorylated activation of its downstream signaling factor IκB and the expression of NF-κB in the nuclear.
