Expressions and significance of Six1 in pancreatic carcinoma tissues

Abstract

OBJECTIVE: To investigate the expression profiling of Six1 in various kinds of human pancreatic cancer cell lines and tissues and their clinical significance.METHODS: Western blotting and immunohistochemical staining analysis were employed to investigate the expression of Six1 in 4 different human pancreatic cancer cell lines and 3T3 cell line,and tissues of pancreatic cancer,benign pancreatic lesions tissues,pancreatitis and normal adjacent pancreatic tissues.RESULTS: The results of Western blotting demonstrated that expression of Six1 was detected in Bxpc-3 and CFPAC-1 cell lines,also,its expression level was significantly increased in pancreatic cancer tissues and benign pancreatic lesions tissues compared with normal adjacent pancreatic tissues.The results of IHC indicated that Six1 expression was positive in 34(85.0%) pancreatic cancer tissues,3(75.0%) benign pancreatic lesions tissues,6(60.0%) pancreatitis and normal adjacent pancreatic tissues.The expression of Six1 had close relation with distant metastasis and tumor size.(P0.05).CONCLUTION: Over-expression of Six1 is the preliminary incidence which would lead to the formation of pancreatic cancer and showed a close relationship with tumor size and distant metastasis.
