Abstract

[Objective]To investigate the effect of TMP on multidrug resistance reversal and P-glycoprotein(P-gp) expression in human breast cancer MCF-7/ ADM cell line.[Methods] The sensitivity of MCF-7/ADM cells and the reversal of resistance were assessed by MTT assay.P-gp expression was determined by flow cytometry.[Results] The non-cytotoxic dose(320g/ml) and low-cytotoxic dose(320g/ml) of TMP decreased significantly the IC_(50) value of ADM to MCF-7/ADM cells(P0.01),and reversed the resistance by 2.14 folds,2.80 folds respectively compared with the IC_(50) value in MCF-7/ADM and reduced P-gp expression from(90.60±0.40)% to(69.10±1.65)% and(60.30±1.25)% respectively.[Conclusion]: TMP could partially reverse the resistance to ADM in human breast cancer MCF-7/ ADM cell line, which may be related to inhibiting P-gp expression in MCF-7/ADM cells.
