Abstract
AIM To study the effect of bumetnide on the contraction of guinea pig tracheal smooth muscle induced by calcium, histamine, acetylcholine and prostaglandin F2α in vitro. METHODS To observe the effect of bumet on cumulative concentration effect curves elicited by Ca~2+ and histamine, and on the contraction of tracheal strips induced by Ach and PGF2α. RESULTS Bumet caused depression of cumulative concentration effect curves elicited by Ca~2+ and histamine with pD′2 value 308±048 and 561±013, repectively. The inhibitory action of bumetnide on Ach 10~-5 mol·L-1 and PGF2α 5×10-3 mol·L-1 was found and the IC50 as (509×10-6±016×10-6) mol·L-1 and (513×10-6±017×10-6) mol·L-1, respectively. CONCLUSION Bumet has inhibitory effect on the contraction of guinea pig tracheal smooth muscle induced by calcium, histamine, acetylcholine and prostaglandin F2α。
