Abstract

AIM To study the effect of bumetnide on the contraction of guinea pig tracheal smooth muscle induced by calcium, histamine, acetylcholine and prostaglandin F2α in vitro. METHODS To observe the effect of bumet on cumulative concentration effect curves elicited by Ca~2+ and histamine, and on the contraction of tracheal strips induced by Ach and PGF2α. RESULTS Bumet caused depression of cumulative concentration effect curves elicited by Ca~2+ and histamine with pD′2 value 308±048 and 561±013, repectively. The inhibitory action of bumetnide on Ach 10~-5 mol·L-1 and PGF2α 5×10-3 mol·L-1 was found and the IC50 as (509×10-6±016×10-6) mol·L-1 and (513×10-6±017×10-6) mol·L-1, respectively. CONCLUSION Bumet has inhibitory effect on the contraction of guinea pig tracheal smooth muscle induced by calcium, histamine, acetylcholine and prostaglandin F2α。
