Abstract

AIM To investigate distribution of polymorphism of Rb gene intron 20 VNTR in the aged and whether there is change between youth and the aged in polymorphism of Rb gene intron20 VNTR, To study whether polymorphism of Rb gene VNTR is associated with cellular aging. METHODSPolymorphism of Rb gene (intron 20) VNTR was examined in 56 cases of unrelated youth and 68 aged people by polymerase chain reaction (PCR). RESULTS There were 10 alleles ranging from 260 bp to 330 bp in youth. Repeat base was 5. The allele frequencies were 0.0009~0.295. The heterozygosity of Rb (intron 20) locus was 0.70. Polymorphism information content (PIC) was 0.86. There were 10 alleles ranging from 240 bp to 390 bp in youth. Distribution of genetic polymorphism of Rb gene (intron 20) VNTR was significantly different between youth and the aged (P0.001), CONCLUSION Change of Polymorphism of Rb gene VNTR may be one of the mechanisms of cellular aging.
