Abstract
Objective To investigate the clinical efficacy of Pemetrexed combined with Cisplatin in the treatment of advanced non-small cell lung cancer.Methods 94 patients with advanced non-small cell lung hospitalized in the Zhumadian Central Hospital from January 2010 to December 2011 were divided into two groups,with 48 patients in the study group and 46 patients in the control group.The control group received 1000 mg/m2 of Gemcitabine on day 1 combined with 25 mg/m2 of Cisplatin from day 1 to day 3.The study group received 500 mg/m2 of Pemetrexed on day 1 combined with 25 mg/m2 of Cisplatin from day 1 to day 3.One treatment cycle consisted of 21 days for both groups.The efficacy,median time to progression(TTP),the median survival time(MST) and the increase rate and decrease rate of life quality of the two groups were observed and compared.The adverse reactions during treatment were ob served.Results The total effective rate of the control group was 71.7% and that of the study group was 83.3%,with statistically significant difference between the two groups(P 0.05).The median progress-free survival time(mPFS) of the control group and the study group was 3.8 months and 5.2 months,with statistically significant difference between the two groups(P 0.05).The median MST of the control group and the study group was 11.2 months and 15.8 months,with statistically significant difference between the two groups(P 0.05).Comparing the increase rate and decrease rate of life quality,the KPS score increased in 18 patients(39.1%),stabilized in 18 patients(39.1%) and de creased in 10 patients(21.7%) in the control group and the KPS score increased in 21 patients(43.8%),stabilized in 22 patients(45.8%) and decreased in 5 patients(10.4%) in the study group,with statistically significant differences between the two groups(P 0.05).The main adverse reactions were myelosuppression,nausea,vomiting,hair loss and whole body fatigue.The study group was lower than the control group in the myelosuppression and the incidence of nausea and vomiting,with statistically significant differences(P 0.05),but similar to the control group in other adverse reactions(P 0.05).Conclusion In the treatment of advanced non-small cell lung cancer,Pemetrexed combined with Cisplatin shows more significant efficacy and fewer toxic reactions than Cisplatin,thus can be used as the firstline treatment medicine for the patients with advanced non-small cell lung cancer.
