Clinicopathological Correlation Research about Expression of CEA mRNA in the Peripheral Blood and Non-Small Cell Lung Cancer

Abstract

Objective To discuss the quantitative detection of CEA mRNA for therapeutic and prognostic significance by analysing clinicopathological correlation of expression of CEA mRNA in the peripheral blood and non-small cell lung cancer.Methods Using carcinoembryonic antigen mRNA(CEA mRNA) as a target gene,CEA mRNA had been examined by reverse real-time quantitative polymerase chain reaction in pretherapy and post-therapy peripheral blood of 49 non-small cell lung cancer patients,10 healthy controls and 10 benign bellows diseases patients.The non-small cell lung cancer patients had been supervised peripheral blood CEA mRNA before and after therapy but the healthy volunteers examined only one time.Results ①There were no peripheral blood CEA mRNA expression in 10 cases healthy volunteers and 10 cases benign bellows diseases patients but non-small cell lung cancer patients had positive rate was 77.6%.CEA mRNA expression was showed in 5 cases non-small cell lung cancer tissues.②The expression rate of CEA mRNA was 42.9%(3/7)、71.4%(5/7)、83.3%(15/18)、88.2%(15/17) in peripheral blood with non-small cell lung cancer of TNM stage Ⅰ、Ⅱ、Ⅲ、Ⅳrespectively.The positive rate increased with the disease evolving.The positive expressionrate was statistically significant between Ⅰ、Ⅱstage and Ⅲ、Ⅳstage(χ2=4.689,P0.05;t=-2.296,P=0.026).③The numerical value of peripheral blood CEA mRNA of the operating patients before and after operation was changed (t=2.267,P=0.033),the expression of CEA mRNA was decline after operation.The numerical value of peripheral blood CEA mRNA of the Ⅲb andⅣstage patients before and after radiotherapy and chemotherapy was changed (P=0.016),the expression of CEA mRNA was decline after radiotherapy and chemotherapy.With the expression of peripheral blood CEA mRNA changed,the clinical curative effect is different (F=0.728,P=0.04;γ=0.629,P=0.01).Conclusion The method of reverse real-time quantitative polymerase chain reaction is sensitive and specific for detection of CEA mRNA expression in the peripheral blood of non-small cell lung cancer patients.Non-small cell lung cancer might be regarded as systemic disease even in early stage.Detecting the expression of CEA mRNA in peripheral blood of the non-small cell lung cancer may contribute to improve decision therapeutic effect and indications for therapy.
