Abstract

[Objective] The propose was to study the factors affecting callus induction of Leonurusn artemisia.[Method] With the true leaves which germinated from L.artemisia seeds as explants and the MS medium as the basic medium,the effects of the plant hormone and the sucrose with different concentration and the different culture methods and illumination conditions on the callus induction rate of L.artemisia were studied.[Result] When 2,4-D was 0.5 or 1.0 mg/L and 6-BA was 2.0 mg/L in the medium,the callus induction rate of L.artemisia were higher,with the highest as 93.3%.The callus induction rate was increased with the increment of sucrose content in the medium,it got highest when the sucrose content reached to 30 g/L and it could be decreased when the sucrose content surpassed 30 g/L.Among 3 kinds of culture modes,the callus induction rate from the solid culture was 1.63 times of that from the liquid culture.Among 3 kinds of illumination,the callus induction rate under whole light could be up to 92.8%,while that under whole dark was only 43.6%.[Conclusion] Adding the rational plant hormone combination and sucrose and adopting the suitable culture mode and the illumination condition could be propitious to the induction of L.artemisia callus.
