Beneficial Effect of Intracellular Trehalose on Viability of Freeze-Dried Red Blood Cells

Abstract

Currently,freeze-drying is one of the best methods for the preservation of red blood cells.The effects of intracellular trehalose on the viability of freeze-dried red blood cells were investigated.The fresh red blood cells were incubated in trehalose solutions of various concentrations at 37℃ for 7 hours.The results show that the intracellular trehalose concentration increased with the increasing concentration of extracellular trehalose solution.The maximum concentration of intracellular trehalose was 35mmol/L in this study.Red blood cells loaded with trehalose were lyophilized with the concentrations of trehalose of 15%,22.5%,30%,and 37.5%,respectively.The results show that the viability of freeze-dried red blood cells increased with the increase of intracellular trehalose concentration.
