Abstract
The factors affecting mesophyll protoplasts isolation from leaves of Arabidopsis thaliana Columbia were reported in this paper.The results showed that the yields of protoplasts were affected by the concentration of celluase and sorbitol,incubation times,the pH of incubational solution.The best yield of mesophyll protoplast were obtained using young leaves incubated in the enzyme solution containing 20 g/L cellulose and 140 g/L sorbitol,at pH 5.8 incubating for 1.5 hours.Moreover,the young leaves had higher yield of mesophyll protoplast compared to older ones.
