Abstract

Objective To investigate the effects and mechanisms of the PI3K/Akt /PTEN pathways on the clear cell renal cell carcinoma. And to provide novel strategy and experimental evidence for clinical therapy for clear cell renal cell carcinoma. Methods 60 cancer tissues and adjacent tissues sections immunohistochemical stained according to routine protocols to evaluate PI3K and Akt protein expression.40 fresh cancer tissues and fresh adjacent tissues reverse transcription polymerase chain reaction(RT-PCR) according to routine protocols to evaluate PI3K,Akt and PTEN mRNA expression. Results PI3K and Akt proteins were expressed at a higher rate in RCC compared to adjacent normal tissue. The rate of positive expression of PI3K and AKT proteins in RCC were positively correlated;PTEN protein was expressed at a higher rate in adjacent normal tissue compared to renal cell carcinoma. Conclusion PI3K and Akt protein abnormal expression suggests that the presence of overall PI3K/Akt signal transduction pathway activated in renal clear cell carcinoma, the tumor suppressor gene PTEN can regulate the activity of the signaling pathway in renal clear cell carcinoma.
