Abstract

For the protoplast isolation of Morchella, the optimal age of mycelia was 3d and 0.6ml/L sucrose was the best osmotic stabilizer. The optimal enzymolysis temperature was 30℃and optimal enzymolysis time was 3h. The highest protoplast yield was 3.35×106/100mg. When 0.6mol/L sucrose was used for stabilizer of the regeneration medium of CYM, the regeneration frequency of protoplast was 0.171%. These results and data would provide important parameters for the strain improvement of Morchella through the protoplast technique.
